Inverse PCR was performed on Drosophila melanogaster strains each of which contains a single EP transposable element insertion. (The generation of these insertion strains is described in Rorth P, Szabo K, Bailey A, Laverty T, Rehm J, Rubin GM, Weigmann K, Milan M, Benes V, Ansorge W, Cohen SM. 1998. Systematic gain-of-function genetics in Drosophila. Development 6:1049-1057.) The resultant fragment for each strain was directly sequenced to determine the genomic sequence at the site of insertion.
