Melioidosis is a life-threatening bacterial infection of public health concern caused by the bacterial agent Burkholderia pseudomallei (B. pseudomallei). Rapid simple, and specific detection of B. pseudomallei will help to prevent and control melioidosis. However, B. pseudomallei has close similarity to its sister species within the same genus, which makes it difficult to identify. In this study, the genomic sequences of 1722 B. pseudomallei strains were analyzed to construct large-scale pan-genomes. Subsequently, the specific sequence tags in core genome sequences were screened. These specific sequence tags effectively distinguish B. pseudomallei from all other closely related species.
