We here firstly assembled a complete genome sequence of the rice reference genome (Nipponbare), named T2T-NIP, derived from a total of 36 Gb Hi-C dataset, 1043 Gb PacBio HIFI reads, 173 Gb Nanopore ONT reads, 23 Gb Illumina Paired-End reads, 11 Gb Illumina Paired-End RNA-Seq reads and 10 Gb Full-length RNA-Seq reads.

By evaluating and verifying the quality of the genome in terms of assembly statistics, gap and assembly errors, gene distribution and segmental repeats, we proved the advantage of this new reference genome on assembly completeness, sequence accuracy and functional elements annotation.
